Biological Activity

MI-2 is a potent inhibitor of the menin-MLL interaction with an IC50 of. By disruptively binding to menin, MI-2 blocks its association with MLL fusion proteins, resulting in the downregulation of critical downstream target genes such as HOXA9 and MEIS1. This inhibition suppresses cellular proliferation, impairs self-renewal, and induces cell differentiation and apoptosis in MLL-rearranged leukemia models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
