Biological Activity

MFIis a small-molecule inhibitor of mitofusin-1 (MFN1) and mitofusin-2 (MFN2). By targeting mitofusins to disrupt tethering and oligomerization, MFIinhibits mitochondrial fusion and decreases the mitochondrial aspect ratio, promoting mitochondrial fragmentation and reducing organelle functionality. Downstream, MFIinduces minority mitochondrial outer membrane permeabilization and caspase-3/activation, enhancing cell death in combination with SMAC mimetics. Additionally, MFIsuppresses mitophagy and mitochondria-ER interactions, sensitizing leukemia cells to BH3 mimetics. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
