Biological Activity

MEN1703 (SEL24) is a potent, orally active dual inhibitor of PIM1 (Kd 2 nM), PIM2 (Kd 2 nM), PIM3 (Kd 3 nM), and FLT3 kinase. By blocking both PIM and FLT3 signaling pathways, it suppresses downstream phosphorylation of STAT5 and 4E-BP1, inhibiting cell proliferation and inducing apoptosis in acute myeloid leukemia models.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
