Biological Activity

Emlenoflast (MCC-7840) is a sulfonylurea derivative that acts as a potent and selective inhibitor of the NLRP3 inflammasome, demonstrating nanomolar potency in human and mouse microglia. Selective inhibition of the NLRP3 inflammasome complex prevents caspase-1 activation and suppresses the downstream maturation and release of proinflammatory cytokines IL-1β and IL-18. This targeted blockade attenuates neuroinflammation and preserves dopaminergic neuronal integrity in preclinical models. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
