Mal-PEG6-NHS ester is a heterobifunctional, non-cleavable antibody-drug conjugate (ADC) linker featuring a maleimide group, a hydrophilic hexameric polyethylene glycol (PEG6) spacer, and an amine-reactive N-hydroxysuccinimide (NHS) ester. The maleimide moiety selectively reacts with free sulfhydryl groups on cysteine residues, while the NHS ester crosslinks primary amine groups on lysine residues or payload molecules. The incorporation of the hydrophilic PEG chain enhances conjugate solubility and reduces plasma aggregation, facilitating stable payload conjugation in ADC development. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
