Biological Activity

MAK683 is a potent inhibitor of embryonic ectoderm development (EED), a core regulatory subunit of Polycomb Repressive Complex 2 (PRC2). By binding to the H3K27me3-binding pocket of EED, MAK683 prevents PRC2 activation and suppresses its methyltransferase activity. This downstream blockade reduces cellular histone H3 lysine 27 trimethylation (H3K27me3) levels, leading to derepression of target genes and inhibition of cell proliferation. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
