M(MSC2504877) is a novel, drug-like small-molecule inhibitor of tankyrase enzymes within the PARP superfamily. Target inhibition leads to suppression of the canonical Wnt/β-catenin signaling pathway in tumor cells. Downstream, this pathway inhibition suppresses Cyclin D2 and Cyclin E2, reduces retinoblastoma protein phosphorylation, and inhibits the growth of APC-mutant colorectal tumor cells by promoting G1 cell cycle arrest and cellular senescence. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
