LY2880070 is an orally active inhibitor of checkpoint kinase 1 (CHK1). Inhibiting CHK1 impairs the cell cycle checkpoint response and destabilizes DNA replication during genomic stress. When combined with the DNA-damaging agent gemcitabine, LY2880070 promotes replication fork instability and elevates markers of replication stress and DNA damage, including pKAP1, pRPA32, and γH2AX, leading to cytotoxic responses in patient-derived organoid models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
