LY345899 is a folate analog that acts as a potent dual inhibitor of methylene tetrahydrofolate dehydrogenase 1 (MTHFD1; IC50 = 96 nM) and methylene tetrahydrofolate dehydrogenase 2 (MTHFD2; IC=. By targeting these enzymes in the folate pathway, it disrupts NADPH and redox homeostasis, leading to cellular death under oxidative stress. Consequently, LY345899 significantly suppresses tumor growth and reduces tumor weight in colorectal cancer patient-derived xenograft models. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
