Biological Activity
LUNA18 (Paluratide) is an orally bioavailable-mer cyclic peptide KRAS inhibitor that binds to KRAS GD with a Kd ofand blocks the KRAS GD–SOS1 interaction with an ICof < 2.2 nM. By disrupting RAS–GEF protein interactions, it suppresses downstream MAPK/ERK and PI3K/AKT signaling, demonstrating antiproliferative activity against KRAS-mutant cancer cell lines, including AsPC-1 pancreatic cancer cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
