Lufotrelvir (PF-07304814) is a phosphate prodrug of the active antiviral compound PF-00835231, functioning as an inhibitor of the SARS-CoV-2 3CL protease (3CLpro, also known as main protease or Mpro). Cleavage of the phosphate group yields PF-00835231, which targets the viral 3CLpro active site, disrupting viral polyprotein processing and suppressing viral replication. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
