Biological Activity

LRK-4189 is an orally active degrader targeting the lipid kinase PIP4K2C. Target-specific depletion of PIP4K2C activates intracellular interferon signaling pathways in microsatellite-stable colorectal cancer models. Consequently, PIP4K2C degradation by LRK-4189 promotes immunogenic tumor cell death and endogenous apoptosis, while effectively sensitizing microsatellite-stable colorectal tumors to natural killer cell killing and dendritic cell phagocytosis to stimulate anti-tumor immune responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
