LP-533401 is a selective inhibitor of tryptophan 5-hydroxylase-1 (TPH1), the rate-limiting enzyme in peripheral serotonin biosynthesis. By inhibiting TPH1, LP-533401 suppresses gut-derived serotonin production without affecting central nervous system serotonin levels. This reduction in peripheral serotonin downstream signaling leads to increased osteoblast proliferation and enhanced bone formation, resulting in restored bone mass in preclinical models of osteoporosis.
