Biological Activity

LJI308 is a potent pan-RSK inhibitor targeting RSK1, RSK2, and RSK3. By blocking p90 ribosomal S6 kinase signaling downstream of the MAPK/ERK pathway, LJI308 attenuates target substrate phosphorylation, including ribosomal protein S6 and glycogen synthase kinase 3 beta. This inhibition interrupts downstream cellular processes, leading to suppressed cell proliferation, survival, and motility in cancer cell models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
