Lefamulin (BC-3781) is a pleuromutilin antibiotic that acts as an inhibitor of bacterial protein synthesis by binding to the peptidyl transferase center of the 50S bacterial ribosomal subunit. By binding to 23S rRNA within the peptidyl transferase center, it blocks peptide bond formation and halts nascent peptide chain elongation. This targeted ribosomal inhibition disrupts bacterial translation, resulting in broad-spectrum antibacterial activity against key respiratory pathogens including Streptococcus pneumoniae and Staphylococcus aureus. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
