Biological Activity

LC-2 is a first-in-class PROTAC degrader targeting endogenous KRAS G12C with DC50 values ranging from 0.25 μM to 0.76 μM. The molecule utilizes an MRTX849 warhead to covalently engage KRAS G12C and recruits the VHL E3 ubiquitin ligase, promoting rapid and sustained ubiquitination and proteasomal degradation of the mutant protein. This targeted degradation effectively suppresses downstream MAPK signaling in KRAS G12C-mutant cancer cells.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
