Larazotide acetate is a synthetic peptide antagonist of zonulin that regulates intestinal tight junction assembly and mucosal barrier function. By blocking zonulin-receptor interactions, it prevents intracellular actin polymerization and the rearrangement of tight junction proteins, including ZO-1 and occludin. Consequently, larazotide acetate maintains paracellular barrier integrity and inhibits pathological increases in intestinal epithelial permeability caused by inflammatory stimuli or gliadin exposure.
