Biological Activity

Laduviglusib trihydrochloride (CHIR-99021 trihydrochloride) is a potent, selective GSK-3 inhibitor targeting GSK-3α and GSK-3β, exhibiting over 500-fold selectivity over CDC2 and ERK2. Through GSK-3 inhibition, it robustly activates the Wnt/β-catenin signaling pathway, promoting the self-renewal of mouse and human embryonic stem cells and driving autophagy. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
