KyA33 is a selective, non-covalent small-molecule inhibitor of ALDH1A2 with dual activity targeting ALDH1A2 and ALDH1A3. By blocking local retinoic acid biosynthesis and suppressing autocrine retinoic acid signaling in both dendritic cells and tumor cells, KyA33 effectively restores dendritic cell function, promotes T cell activation, and enhances localized antitumor immunity within the tumor microenvironment.
