KU-0060648 is a potent dual inhibitor of DNA-dependent protein kinase (DNA-PK) and phosphatidylinositol 3-kinase (PI3K) isoforms. It inhibits downstream AKT/mTOR signaling and DNA repair pathways, impairing double-strand break repair and cell survival mechanisms. Consequently, KU-0060648 suppresses cancer cell proliferation, induces cell cycle arrest and apoptosis, and sensitizes malignant cells to DNA-damaging agents and ionizing radiation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
