Biological Activity
KT-333 is a potent and selective heterobifunctional degrader targeting signal transducer and activator of transcription 3 (STAT3) by recruiting the VHL E3 ligase. Targeted degradation of STAT3 downregulates downstream anti-apoptotic effectors such as MCL1, thereby suppressing survival signaling pathways. Consequently, KT-333 induces mitochondrial dysfunction and apoptosis, demonstrating robust antitumor activity in preclinical models of ALK-positive anaplastic large cell lymphoma and acute myeloid leukemia. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
