Padnarsertib (KPT-9274) is an orally bioavailable, non-competitive dual inhibitor targeting p21-activated kinase 4 (PAK4) and nicotinamide phosphoribosyltransferase (NAMPT, IC50 ~120 nM in cell-free enzymatic assays). Inhibition of PAK4 and NAMPT depletes intracellular NAD+ and ATP levels, suppressing downstream Wnt/β-catenin signaling pathways. Consequently, Padnarsertib disrupts cell cycle dynamics, inhibits tumor cell proliferation, and induces apoptosis across various preclinical cancer models.
