Ko143 is a potent and selective inhibitor of the multidrug transporter ATP-binding cassette sub-family G member 2 (ABCG2/BCRP). By blocking ABCG2-mediated transport and ATPase activity, Ko143 prevents the active efflux of diverse therapeutic compounds across cell membranes. Downstream, this inhibition increases intracellular substrate accumulation and sensitizes drug-resistant cancer cells to ABCG2-transported chemotherapeutic agents, reversing multidrug resistance phenotypes.
