Biological Activity

KO-947 is a potent and selective inhibitor of ERK1/2 kinases that suppresses the MAPK signaling pathway. By directly blocking ERK1 and ERK2 catalytic activity, KO-947 disrupts downstream oncogenic signal transduction essential for tumor cell survival and proliferation. In cancer models driven by dysregulated MAPK signaling, KO-947 effectively inhibits cellular proliferation at low nanomolar concentrations. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
