Biological Activity

KN-93 hydrochloride is a methoxybenzenesulfonamide derivative and inhibitor of Ca2+/calmodulin-dependent protein kinase II (CaMKII) with a Ki of 0.37 µM. By suppressing CaMKII activity, it prevents Ca2+-mediated phosphorylation of tyrosine hydroxylase (TH) and reduces TH reaction rates. This downstream inhibition of TH phosphorylation ultimately decreases intracellular dopamine production, demonstrating its utility in modulating Ca2+/calmodulin-dependent signaling pathways. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
