Biological Activity

KN-62 is a potent inhibitor of Ca2+/calmodulin-dependent protein kinase II (CaMKII, and a non-competitive antagonist of purinergic receptor P2RX7. It displays high selectivity for CaMKII relative to PKA, PKC, and MLCK. By inhibiting CaMKII and P2RX7 pathways, KN-62 regulates intracellular calcium signaling and suppresses downstream phosphorylation events and inflammatory phenotypic responses. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
