Biological Activity

KL-11743 is a potent, glucose-competitive inhibitor of class I glucose transporters, including GLUT1, GLUT2, and GLUT3. By blocking cellular glucose metabolism, KL-11743 triggers an acute collapse in cellular NADH pools and a striking accumulation of aspartate, driving a metabolic shift toward mitochondrial oxidative phosphorylation. Consequently, inhibition of glucose transport with KL-11743 induces synthetic lethality when combined with mitochondrial electron transport chain inhibitors or in tricarboxylic acid cycle-deficient tumors. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
