Biological Activity
is a potent and selective inhibitor of the KEAP1/NRF2 protein-protein interaction. By disrupting the binding of KEAP1 to NRF2, KI696 prevents KEAP1-mediated degradation of NRF2, leading to NRF2 accumulation and nuclear translocation. This activation upregulates downstream antioxidant genes including Nqo1, Ho-1, Txnrd1, Srxn1, Gsta, and Gclc in a dose-dependent manner to enhance cytoprotective cellular defenses against oxidative stress. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
