Biological Activity

K-Ras(G12C) inhibitor 12 is a selective allosteric inhibitor targeting mutant K-Ras(G12C). By covalently binding to the cysteine 12 residue within the switch II pocket, the compound locks K-Ras in an inactive GDP-bound state and impairs effector interactions. This allosteric inhibition blocks downstream MAPK/ERK signaling, suppressing cell proliferation and inducing apoptosis in K-Ras(G12C)-mutated cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
