Biological Activity

JY-2 is a selective and orally active inhibitor of the transcription factor forkhead box O1 (FoxO1) that also exerts moderate inhibitory effects on FoxO3a and FoxO4. By suppressing FoxO1-driven transcriptional activity, JY-2 attenuates downstream gluconeogenic gene expression and metabolic signaling pathways. Consequently, targeted inhibition of FoxO1 produces therapeutic anti-diabetic efficacy across cellular and preclinical metabolic disease phenotypes. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
