Biological Activity

JWZ-8-103 is an orally bioavailable molecular glue targeting TRIM21, an E3 ubiquitin ligase that recruits intracellular targets for proteasomal degradation. It promotes the interaction between the TRIM21 PRYSPRY domain and NUP98 to induce selective degradation of the nuclear pore complex. This disruption impairs nuclear pore stability, leading to potent antitumor activity in pancreatic cancer xenografts and patient-derived organoids. [1]
