Biological Activity

JW642 is a potent small-molecule inhibitor of monoacylglycerol lipase (MAGL). By inactivating MAGL, JW642 prevents the degradation of the primary endocannabinoid 2-arachidonoylglycerol (2-AG) in brain tissues. This inhibition leads to accumulation of 2-AG, enhancing cannabinoid receptor-mediated signaling pathways. Consequently, MAGL blockade modulates central nervous system endocannabinoid tone, attenuating pro-inflammatory arachidonic acid pools, dampening neuroinflammation, and inducing downstream antinociceptive responses in neural networks. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
