(+)-JQ1 carboxylic acid is a functionalized derivative of the BET bromodomain inhibitor (+)-JQ1 containing a terminal carboxylic acid moiety. It serves as a key chemical precursor and recruiting element for the synthesis of proteolysis targeting chimeras (PROTACs) targeting BET bromodomains. When conjugated to linkers and E3 ligase ligands, JQ1 derivatives enable targeted ubiquitin-proteasome degradation of BET proteins such as BRD4, resulting in suppressed oncogenic transcription and inhibition of cancer cell proliferation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
