JNK-IN-8 is a potent, irreversible inhibitor targeting JNK1, JNK2, and JNK3 (IC50 1 nM). By covalently modifying a conserved cysteine residue within the kinase ATP-binding pocket, it selectively blocks JNK activity and suppresses downstream phosphorylation of c-Jun. Consequently, JNK-IN-8 inhibits MAPK pathway signaling, leading to reduced expression of AP-1-dependent target genes and altered cell survival responses under cellular stress. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
