JNJ-7777120 is a potent and selective non-imidazole histamine H4 receptor antagonist with a Ki of 4.5 nM, exhibiting >1000-fold selectivity over other histamine receptor subtypes. By inhibiting histamine H4 receptor signaling, it blocks histamine-induced calcium influx and chemotaxis of immune cells such as mast cells and eosinophils. Consequently, JNJ-7777120 demonstrates marked anti-inflammatory and immunomodulatory activity in animal models of allergic disease and inflammation. [1]
