Biological Activity

JNJ-42226314 is a potent, selective, and reversible noncovalent inhibitor of monoacylglycerol lipase (MAGL). By suppressing MAGL-mediated degradation of endocannabinoids, this compound effectively elevates endogenous 2-arachidonoylglycerol (2-AG) levels in tissue and indirectly drives cannabinoid CB1 receptor occupancy. Downstream, JNJ-42226314 demonstrates robust antinociceptive efficacy in rodent preclinical models of both inflammatory and neuropathic pain. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
