Biological Activity

JNJ 303 is a potent inhibitor of the voltage-dependent IKs channel encoded by the KCNQ1/KCNE1 potassium channel complex, displaying an IC50 of 64 nM. Blockade of this repolarizing current impairs cardiac action potential duration and delays ventricular repolarization, resulting in marked QT interval prolongation. Consequently, JNJ 303 disrupts normal cardiac electrophysiology and predisposes ventricular tissue to unprovoked torsades de pointes.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
