JHU395 is an orally bioavailable glutamine antagonist prodrug designed to selectively deliver active glutamine antagonists to target tissues while remaining inert in plasma. By inhibiting glutamine-utilizing metabolic pathways, JHU395 disrupts cellular nutrient utilization and energy production in glutamine-dependent tumor cells. This metabolic blockade impairs cell proliferation and suppresses tumor growth in malignant peripheral nerve sheath tumor models without inducing significant systemic toxicity. [1]
