Biological Activity

JH-RE-06 is a potent REV1-REV7 interface inhibitor with an IC50 of 0.78 μM and a Kd of 0.42 μM for REV1 binding, which disrupts REV1-POL ζ-mediated mutagenic translesion synthesis. By blocking this protein-protein interaction, JH-RE-06 inhibits error-prone DNA lesion bypass, suppresses chemotherapy-induced mutagenesis, and enhances the cytotoxic effects of DNA-damaging agents such as cisplatin in cancer cell lines and animal models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
