Biological Activity

Ivacaftor (VX-770) is a selective CFTR potentiator targeting G551D-CFTR (EC50 of 100 nM) and F508del-CFTR (EC50 of 25 nM). It increases CFTR channel open probability to enhance chloride ion secretion across airway epithelial cells. This process attenuates excessive sodium and fluid absorption to prevent surface dehydration and restore cilia beating frequency in cystic fibrosis models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
