IU1 is a cell-permeable, reversible, and selective inhibitor of USP14 (IC50 =, exhibiting 25-fold selectivity over IsoT. By inhibiting the deubiquitinating activity of USP14 associated with the 26S proteasome, IU1 promotes the degradation of specific proteasomal substrates and regulates cellular proteostasis. Additionally, USP14 inhibition by IU1 modulates autophagic flux and induces autophagy in various cellular models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
