Biological Activity

ITI-214 is a potent, CNS-penetrant, and orally bioavailable phosphodiesterase-1 (PDE1) inhibitor that targets human PDE1A, PDE1B, and PDE1C isoforms. By inhibiting PDE1 enzymatic activity, ITI-214 blocks the hydrolysis of cyclic nucleotides, thereby elevating intracellular cAMP and cGMP levels to modulate downstream signaling cascades. These pathway alterations enhance neuronal function and offer potential therapeutic benefits for cognitive impairment associated with neurodegenerative and neuropsychiatric disorders.[1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
