Biological Activity

ITF3756 is a selective histone deacetylase 6 (HDAC6) inhibitor bearing a pentaheterocyclic scaffold. By dampening TNF-α pathway activation, ITF3756 downregulates PD-L1 expression and upregulates costimulatory CD40 expression in activated monocytes, promoting a less immunosuppressive phenotype and enhancing T-cell proliferation. In vivo, ITF3756 reduces tumor growth in murine colon cancer models, supporting its utility in cancer immunotherapy research. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
