IRG1-IN-1 is a potent inhibitor of immune-responsive gene 1 (IRG1), an enzyme that regulates immunometabolism in macrophages. By targeting IRG1 activity, it blocks macrophage-mediated metabolic crosstalk in the tumor microenvironment and reverses tumor-associated macrophage-induced resistance to PARP inhibitors. Consequently, IRG1-IN-1 enhances PARP inhibitor sensitivity to suppress tumor growth in immunocompetent models of BRCA1-deficient breast cancer. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
