Biological Activity

IRAK4-IN-4 is a potent inhibitor of interleukin-1 receptor-associated kinase 4 (IRAK4) and cyclic GMP-AMP synthase (cGAS). Inhibition of IRAK4 suppresses downstream Toll-like receptor and interleukin-1 receptor signaling pathways, leading to the inhibition of NF-kB activation and reduced expression of pro-inflammatory cytokines. These cellular effects diminish inflammatory responses and attenuate immune cell activation associated with autoimmune disorders and inflammatory conditions. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
