IRAK4-IN-2 is a reversible kinase inhibitor targeting interleukin-1 receptor-associated kinase 4 (IRAK4). By modulating IRAK4 activity, it impairs signal transduction downstream of both Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) pathways. This inhibition blocks the activation of downstream NF-kB and MAPK signaling cascades, resulting in the suppressed expression of pro-inflammatory cytokines and attenuated immune responses. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
