Biological Activity

IOX4 is a potent and selective inhibitor of hypoxia-inducible factor prolyl-hydroxylase 2 (PHD2). By inhibiting PHD2 activity, IOX4 prevents the proline hydroxylation and subsequent degradation of hypoxia-inducible factor alpha (HIFα), leading to HIFα accumulation across various tissue types and human cell lines. This activation of the HIF signaling pathway regulates cellular responses to hypoxia and downstream transcription.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
