INT-777 is a potent and selective agonist of the G protein-coupled bile acid receptor TGR5 (GPCR19). Activation of TGR5 by INT-777 stimulates intracellular cAMP accumulation, promoting glucagon-like peptide-1 (GLP-1) secretion in enteroendocrine cells. Downstream signaling enhances mitochondrial function and oxidative energy expenditure in brown adipose tissue and skeletal muscle, contributing to improved glucose tolerance and metabolic homeostasis in vivo.
