Biological Activity

Rentosertib (INS018-055) is a small-molecule inhibitor of TRAF2- and NCK-interacting kinase (TNIK) and MAPKBy targeting TNIK, the compound attenuates downstream pro-fibrotic pathways including Wnt and TGF-β signaling, thereby repressing transcriptomic programs driving tissue remodeling. Consequently, TNIK inhibition reduces myofibroblast activation, decreases extracellular matrix deposition, and suppresses inflammatory signaling to exert anti-fibrotic phenotypes across preclinical disease models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
