Biological Activity
Inixaciclib (NUV-422) is a potent inhibitor of CDK2, CDK4, and CDK6. It blocks cyclin-dependent kinase activity in the G1/S cell cycle transition pathway, impairing retinoblastoma protein phosphorylation and halting tumor cell proliferation. In preclinical evaluation, inixaciclib suppresses glioma cell line growth in vitro and demonstrates antitumor efficacy in glioblastoma xenografts, CDK4/6 inhibitor-resistant metastatic breast cancer models, and anti-androgen-resistant prostate cancer models.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
